Abstract:Objective To establish a UPLC-MS/MS method for the simultaneous determination of astragaloside IV, verbascoside, hesperidin, ferulic acid, glycyrrhizic acid, and liquiritin in Buzhong Yiqi Pills(water pills). Methods Agilent Zorbax Eclipse XDB-C18 column(100 mm × 2.1 mm, 1.8 μm) was used, and the mobile phase was 5 mmol/L ammonium acetate solution-acetonitrile (containing 0.1% formic acid), gradient elution; the ionization mode of mass spectrometry was electrospray source (ESI), and the quantitative analysis was carried out by using multiple reactive ion monitoring mode (MRM). Results Each compound had a good linear relationship in the corresponding concentration range, the r2 was between 0.994~0.999, the recovery rate of standard addition ranged from 86.7% to 103.5%, and the corresponding RSD was 3.8% to 7.6%. Conclusion This method is accurate and reliable qualitatively and quantitatively, with good repeatability, small matrix interference and high sensitivity, which meets the requirements of measuring the content of 6 components in Buzhong Yiqi Pills (water pills).
