    摘要:

    目的 研究加味楂曲饮(JiaWeiZhaQuYin,JWZQY)防治非酒精性脂肪肝病(Non-Alcoholic Fatty Liver Disease,NAFLD)的药效学。方法 SDF级大鼠随机分为6组:空白组、模型组、对照组、低剂量组、中剂量组、高剂量组。空白组给予基础饲料,其余各组给予高脂饲料造模,自造模第1天起,空白组、模型组大鼠灌服等体积生理盐水,对照组灌服等体积复方蛋氨酸胆碱混悬液(0.038g/mL),低、中、高剂量组分别灌服等体积JWZQY混悬液(JWZQY浓度相应为1g/mL、2g/mL、4g/mL),8周后,常规油红O法(改良Lillie)观察肝脏组织形态学改变,全自动生化仪测定血清丙氨酸基转移酶(Alanine Transaminase,ALT)、谷氨酸基转移酶(Aspartate Amino Transferase,AST)、甘油三醋(Triglycerides,TG)、总胆固醇(Total Cholesterol,TC)水平,比色法检测血清游离脂肪酸(Free Fatty Acid,FFA)水平。结果 高、中剂量JWZQY治疗后肝脏脂肪变程度明显减轻(P<0.05);与模型组相比,对照组及高、中剂量组ALT、AST、TC、TG水平降低,差异有统计学意义(P<0.01)。与对照组比较,高剂量ALT、AST、TC、TG水平降低,差异有统计学意义(P<0.01)。与模型组相比,中剂量组血清中FFA减少,差异有统计学意义(P<0.01)。结论 JWZQY可有效防治实验性非酒精性脂肪肝,减少FFA生成,具有良好研究前景。

    Abstract:

    Objective To study the pharmacodynamics of Jiawei ZhaQuYin(JWZQY)interventing non-alcoholic fatty liver. Methods SDF level rats were randomly divided into six groups:blank group,model group,positive control group,low-dose group,middle dose group and high dose group. The blank group was given basic feed,the rest of the group was given a high fat diet model,Since the Modeling day,the blank and model rats fed with an equal volume of saline,the positive control group was given the same volume of compound methionine choline solution(0. 038g/ml)orally. Low,medium and high dose group was given the same volume of a solution containing 1g/ml,2g/ml,4g/ml concentration JWZQY accordingly. After 8 weeks,the conventional oil red O method(modified Lillie)was used to observe the morphological changes of the liver tissue,The automatic biochemical analyzer used to measure the level of serum Alanine transaminase,Aspartate Amino Transferase,Triglyceride,Total Cholesterol,Elisa was used to detect the levels of Free Fatty Acid. Results Through light microscope,we can see that the high-fat diet led to liver cells diffuse enlargement and round in rat the hepatic lobule,the sinusoidal smaller even not visible,the cytoplasm contains a large number of red dye fat droplets of varying sizes,showing steatosis. High-dose and medium-dose JWZQY can significantly reduce hepatic steatosis(P<0. 05). Compared with the model group,The of level serum ALT,AST,TC,TG of the positive control rat was reduced,the difference was statistically significant(P<0. 01),difference of serum ALT,AST,TC,TG level in low-dose group not statistically significant(P>0. 05),Low,medium and high dose group,the serum ALT,AST,TC,TG level was significantly lower,The difference was statistically significant(P<0. 01). Compared with the positive control group,serum ALT,AST,TC,TG level is reduced in high dose group,the difference was statistically significant(P<0. 01),but in the medium dose group and high dose group,serum ALT,AST,TC,TG no statistically significant difference(P>0. 05). Compared with model group,the difference of serum FFA in medium dose group was statistically significant(P<0. 01),low-dose serum FFA was no significant difference (P>0. 05). Conclusion JWZQY can effectively intervene in the experimental non-alcoholic fatty liver,reduce FFA generation,having good market prospects.
