    摘要:

    目的观察葛根素(Pue)对IL-1β损伤大鼠关节软骨细胞核因子-κB(NF-κB)的影响。方法取10日龄大鼠关节软骨作体外细胞培养,培养后软骨细胞随机分为正常组、IL-1β组、地塞米松组、Pue50组、Pue100组、Pue200组等6组。后5组分别给药后24h,用实时定量PCR法检测各组NF-κBp65的表达。结果IL-1β损伤软骨细胞核内NF-κBp65的表达明显增强,后4组与IL-1β组比较差异有统计学意义(P<0.05),且随着剂量上升(50μmol/L、100 μmol/L、200 μmol/L)使其NF-κB的表达量逐步下降。结论葛根素具有一定的抑制NF-κBp65活化的作用,为防治骨性关节炎提供新的途径。

    Abstract:

    Objective To study the influence of Puerarin(Pue) on Nuclear factor-kappaB(NF-κB) of rat articular chondrocytes injuried by IL-1β. Methods The chondrocytes in vitro were got from articular cartilage of ten-day old rat. Cultured chondrocytes were randomly divided into six groups including normal group,IL-1β group,dexamethasone group,Pue 50 group,Pue 100 group,Pue 200 group. The last five groups were treated with 10μmol IL-1β(5 μg/L),IL-1β(5 μg/L) 10 μmol+Dexamethasone 10-9 mol,IL-1β (5 μg/L) 10 μmol+Pue 50 μmol/L,IL-1β (5 μg/L)10μmol+Pue 100 μmol/L,IL-1β(5 μg/L) 10 μmol+Pue 200 μmol/L. The expressions of NF-κBp65 were assessed by Real-time quantitative Polymerase Chain Reaction. Results The expressions of NF-κBp65 were increasd obviously in damaged chondrocytes nucleus. The last four groups were superior to the group of IL-1β(P<0. 05)severally. And with the dose increased(50 μmol/L,100 μmol/L,200 μmol/L) the expressions of NF-κBp65 were rising gradually. Conclusion Pue can markedly inhibit the activities of NF-κBp65,and it maybe one of the new method of prevention and treatment of osteoarthritis.
