Effects of Linggui Zhugan Decoction on Hemodynamics and BNP in Serum of Model Rats with Ventricular Remodeling after Acute Myocardial Infarction

    摘要:

    目的观察苓桂术甘汤对急性心肌梗死后心室重构模型大鼠心功能的影响及各组大鼠血清脑钠肽含量的变化,探讨苓桂术甘汤干预心室重构的机制。方法采用冠状动脉结扎法复制急性心肌梗死大鼠模型,造模2周后将模型大鼠随机分为假手术组,模型组,卡托普利组,苓桂术甘汤低、中、高剂量组。连续给药4周,观察各组大鼠血流动力学参数的变化,采用ELISA法检测血清BNP含量。结果连续给药4周后,各组大鼠血流动力学各项指标显著改善,苓桂术甘汤小、中、大剂量组大鼠LVSP、+dp/dtmax和-dp/dtmax均显著上升,LVEDP显著下降,大鼠血清BNP含量显著降低(P<0.01或P<0.05)。结论苓桂术甘汤对急性心肌梗死后心室重构大鼠心脏舒缩性能具有显著的改善作用,其作用与降低大鼠血清BNP含量密切相关。

    Abstract:

    Objective To study the effects of Linggui Zhugan Decoction(LGZGD)interference on ventricular remodeling(VR)in rats after acute myocardial infarction(AMI), through observing hemodynamics and the expression of BNP in Serum. Methods The AMI model was induced by ligation of coronary artery. 2 weeks after the modeling, the rats were randomly classified into Sham group, model, captopril, and low, middle and high dosage of LGZGD group. Sham group and the other five groups were ig administered once daily for consecutive 4 weeks. Then the hemodynamic parameters were observed and the levels of BNP in serum were detected by ELISA. Results The low, middle and high dose of Linggui Zhugan Decoction all significantly increased left ventricular systolic pressure(LVSP), maximal rate of left ventricularpressure of development(+dp/dtmax)and maximal rate of left ventricular pressure of decline(-dp/dtmax)(P<0.05, or P<0.01), and decreased left ventricular end diastolic pressure(LVEDP), and the levels of BNP were significantly decreased(P<0.05, or P<0.01). Conclusion Linggui Zhugan Decoction has marked effects on improving the systolic and diastolic functions of the heart in rats with VR of post-AMI;which might be related to the inhibition of BNP.
