    摘要:

    目的探讨加味四逆汤对刀豆蛋白A(Con A)诱导肝细胞损伤过程中肝脏组织Fas抗原的表达和血清肿瘤坏死因子-α(TNF-α)水平的干预情况。方法雌雄各半的NIH小鼠随机分成5组:正常对照组、模型组、阳性药物组、加味四逆汤大、小剂量组,采用Con A(18mg·kg-1)尾静脉注射制作急性免疫性肝损伤模型。造模给药结束后,观察血清中TNF-α含量变化及肝组织细胞Fas抗原表达。结果模型组小鼠肝脏细胞内有大量的Fas抗原表达,且血清中TNF-α含量也明显升高,与对照组比较差异具有统计学意义(P<0.05);对照组和药物组血清中TNF-α水平变化不大,肝细胞Fas抗原表达不明显。结论加味四逆汤能够干预Con A小鼠肝脏组织Fas抗原和血清TNF-α水平的表达,对急性肝损伤有较好的保护作用。

    Abstract:

    Objective To investigate the expression of Fas antigen in hepatocyte and determination the serum tumor necrosis factor alpha(TNF-α) of Modified Sini Decoction on Hepatocyte Apoptosis induced by Concanavalin A(Con A). Methods The mouse were randomly divided into five groups:normal control group, model group, positive drug group, flavored SND large and small dose group, ALL groups, except for the NS group, were given Con A aqueous solution(18mg. kg-1) via tail vein injection, determination the levers of the serum tumor necrosis factor alpha(TNF-α), and the livers were taken for for the histopathology examination, meanwhile detection of Fas antigen expressions in liver tissue sections. Results The model mice liver cells express large amounts of antigen Fas, and serum TNF-α levels also increased significantly with statistical significance(P<0. 05) difference compared with the control group;TNF drug group and the control group in serum -α levels changed little, Fas antigen expression in liver cells is not obvious. Conclusion Modified Sini decoction has protecting effect on immune Hepatocyte Apoptosis induced by Con A in mice, and the mechanism is probably to anti-Fas antigen and TNF-α.
