    摘要:

    目的研究山茱萸总萜调控伴胰岛素抵抗的PCOS大鼠及其卵巢颗粒细胞分泌作用的机制。方法22日龄雌性SD大鼠皮下注射硫酸普拉睾酮钠9 mg·100g-1·d-1共20 d,伴高脂饮食喂养60 d建立伴IR的PCOS大鼠模型,将PCOS大鼠随机分为模型组和治疗组。治疗组以山茱萸总萜、氯米芬和罗格列酮干预。葡萄糖氧化酶法测定空腹血糖(FBG)。放射免疫法测定空腹胰岛素(Fins)、睾酮(T)、雌二醇(E2)、促卵泡刺激素(FSH)和黄体生成素(LH)的含量。同时,随机取各组大鼠2只皮下注射PMSG 10 IU/kg,48 h后颈椎脱臼法处死。在无菌条件下迅速剖取卵巢,刺破卵泡,获取卵巢颗粒细胞。光镜观察卵巢形态及颗粒细胞分布,用不同浓度山茱萸总萜、氯米芬和罗格列酮干预培养,检测颗粒细胞对葡萄糖的摄取量和分泌孕激素(P)的量。结果与模型组比较,治疗组大鼠Fins浓度均显著降低(P<0.01),ISI显著升高(P<0.01),T和E2浓度均显著降低(P<0.01),FSH浓度显著升高(P<0.01),大鼠卵巢卵泡内颗粒细胞层数明显增加,孕激素分泌增加(P<0.05),葡萄糖摄取能力增加(P<0.05)。与氯米芬和罗格列酮双阳性对照组相比,无明显区别(P>0.05,P>0.05)。结论山茱萸总萜可改善胰岛素抵抗,细胞葡萄糖摄取,并增加其雌激素和孕酮的分泌。

    Abstract:

    Objective To study the dogwood total terpene regulation of PCOS rats with insulin resistance and its mechanism of ovarian granular cells secrete. Methods Female SD rats subcutaneously age of 22, prasterone sulfate sodium 9 mg·100·d-1·g-1, a total of 20 days, 60 days with high fat diet feeding PCOS rats model with IR, PCOS rats were randomly divided into model group and treatment group. The treatment group with total terpene dogwood, chlorine meters Finn and rosiglitazone intervention. Glucose oxidase method determination of fasting blood glucose(FBG). Ria method fasting insulin(Fins), testosterone(T), estradiol(E2), promote the follicle-stimulating hormone(FSH) and luteinizing hormone(LH) content. At the same time, the random take each group two rats subcutaneously PMSG iu/kg, 10 cervical dislocation method executed after 48 h. Under aseptic conditions quickly profile control in the ovary, Pierce follicle, get ovarian granulosa cells. Observed ovarian morphology and granular cell distribution, chloride with dogwood total terpene and m intervention training, detection of granulosa cells to glucose intake and the amount of secretion and progesterone(P). Results Compared with model way, the treatment group rats Fins concentrations were significantly lower(P<0.01), ISI significantly increased(P<0.01), T and E2 concentrations were significantly lower(P<0.01), FSH concentration significantly increased(P<0.01), layer number of particles in rat ovarian follicle cells increased significantly, progesterone secretion increased(P<0.05), the ability of glucose uptake increased(P<0.05). With chlorine meters compared with rosiglitazone double positive control group, no significant difference(P>0.05, P>0.05). Conclusion Total terpene dogwood can improve insulin resistance, cell glucose uptake, and increase the production of estrogen and progesterone.
