    摘要:

    目的改进白及薄层鉴别方法和建立白及的高效液相指纹图谱,为其质量控制提供方法。方法采用甲醇超声提取制备供试品溶液,以三氯甲烷-甲醇-水(6∶0.5∶0.05)为展开剂,1%香草醛硫酸乙醇液为显色剂;采用HPLC法,以Thermo Acclaim 120 C18(4.6mm×250mm,5μm)为色谱柱,乙腈和水为流动相,梯度洗脱,流速为1.0mL/min,检测波长为270nm,柱温为25℃;采用中药色谱指纹图谱评价系统进行相似度分析。结果采用改进后的方法进行白及薄层鉴别,色谱分离效果理想,斑点清晰;建立了白及药材HPLC指纹图谱,确定11个共有峰,不同批次白及特征图谱相似度均在0.95以上。结论该方法简单、可靠,可用于白及药材的鉴别和质量控制。

    Abstract:

    Objective To improve the pharmacopoeia TLC method and establish the HPLC fingerprints of chemical constituents in Bletilla striata to provide the basis for its quality control. Methods The samples of Bletilla striata were treated with the ultrasonic extracting. A mixed solution containing CHCl3-MeOH-H2O(6∶0.5∶0.05) was used as developing agent with ascending manner, and citronellal was used as the color reagent. Thermo Acclaim 120 C18 column(250mm×4.6mm, 5μm) was used with acetonitrile and water as mobile phase in a gradient mode at the flow rate of 1. 0mL/min. The detection wavelength was 270 nm and the temperature of column was set at 25℃. Similarity evaluation system for chromatographic fingerprint of TCM was applied to analysis different batches of Bletilla striata. samples. Results The improved TLC method had the advantages of easy operation, good isolation effects and coloration effects. The HPLC characteristic fingerprint of Bletilla striata has been established. Eleven mutual peaks were selected as the fingerprint peaks in 10 batches sample and similarities were all above 0.95. Conclusion This method is easy and reliable which can be used to judge the habitat and control the quality of Bletilla striata.
