    摘要:

    目的建立百蕊草中百蕊草素Ⅰ、百蕊草素Ⅱ与山奈酚的一测多评含量测定方法。方法采用C18色谱柱(phenomenex,250×4.6mm,5μm);以甲醇-0.4%磷酸溶液为流动相;梯度洗脱(0→5→10→20→45min,45%→50%→55%→60%甲醇);波长365nm;流速1.0mL/min;柱温40℃。以山奈酚为参照物,建立其与百蕊草素Ⅰ和百蕊草素Ⅱ的相对校正因子,并计算其含量,实现一测多评。同时采用外标法测定3个成分的含量,并比较二者差异,以验证一测多评法的准确性和可行性。结果采用一测多评法和外标法测定的含量间无显著差异,RSD<2%. 结论本文建立的一测多评法控制百蕊草的质量可行、准确。

    Abstract:

    Objective To develop a method of quantitative analysis of multi-components by single marker(QAMS) for simultaneously determining kaempferol-3-O-glucorhamnoside, astragalin and kaempferol in Thesium. Methods The chromatographic separation was achieved on C18 chromatographic column(phenomenex, 250×4.6mm, 5μm), the mobile phase was methanol -0.4% phosphoric acid aqueous solution with gradient elution(0→5→10→20→45min, 45%→50%→55%→60% methanol). The wavelength was 365nm. The flow rate was 1. 0 mL/min and the column temperature was 40℃. Kaempferol was used as the reference, the calibration factor of kaempferol-3-O-glucorhamnoside and Astragalin to that of kaempferol were calculated. And the contents of active constituents were calculated by the relative calibration factor to realize QAMS. At the same time, external standard method was used to detect the contents of the three constituents, the two methods were compared to validate the accuracy and feasibility of the new method. Results No significant differences were found in the quantitative results between the measured value and the calculated value. Conclusion The QAMS is feasible and accurate to evaluate the contents of the three active constituents in Thesium.
