    摘要:

    目的建立八角莲TLC鉴别方法和HPLC指纹图谱,为其质量控制提供专属性更好的测试方法。方法采用乙酸乙酯超声提取制备供试品溶液,以氯仿-甲醇-水(9∶1∶0.1)作为展开系统,展距13cm,10%硫酸乙醇溶液为显色剂;采用HPLC法,Thermo AcclaimTM 120 C18(4.6×250 mm,5 μm)色谱柱,乙腈-0.05%磷酸水溶液为流动相,梯度洗脱,流速为1.0mL/min,检测波长为203nm,柱温36℃;采用中药色谱指纹图谱评价系统进行相似度分析。结果建立了八角莲药材的TLC鉴别方法和HPLC指纹图谱,确定11个共有色谱峰,方法学验证结果符合指纹图谱相关要求。结论该方法简单、可靠,可用于八角莲药材的鉴别和质量控制。

    Abstract:

    Objective To establish the TLC identification method and HPLC fingerprint of Dysosma versipellis to provide better specificity test method for the quality control. Methods The sample solution was prepared by ultrasonic extraction with ethyl acetate, and the chloroform-methanol-water(9∶1∶0.1)was used as an expansion system, and the development of 13cm, 10% sulfuric acid ethanol solution was used as the chromogenic reagent;Thermo AcclaimTM 120 C18(4.6×250 mm, 5μm)was used with acetonitrile-0.05% phosphoric acid water solution as mobile phase in a gradient mode at the flow rate of 1. 0mL/min. The detection wavelength was 203 nm and the temperature of column was set at 36℃. Similarity evaluation system for chromatographic fingerprint of TCM was applied to analysis different batches of Dysosma versipellis samples. Results The TLC identification method and HPLC fingerprint of Dysosma versipellis was established, eleven mutual peaks which separation in good condition were selected as the fingerprint peaks in 10 batches sample. Conclusion This method is simple and reliable, can be used for the identification and quality control of Dysosma versipellis.
