    摘要:

    目的〓研究肺癌微环境脾气虚证肿瘤组织相关巨噬细胞microRNA的表达及加味黄芪建中汤的影响。方法〓50只小鼠采用苦寒泻下,劳倦,节食的方法造模,在建立小鼠脾气虚证Lewis肺癌模型后,加味黄芪建中汤灌胃治疗10d,脱颈椎处死小鼠,取肿瘤组织。采用microRNA基因芯片检测正常小鼠巨噬细胞与3组肿瘤组织相关巨噬细胞MicroRNA基因表达的变化。结果〓“脾气虚”组肿瘤组织相关巨噬细胞microRNA的表达谱表达异常(与正常组比较),不同于肿瘤组,具有独特的microRNA变化,而加味黄芪建中汤可明显双向调节“脾气虚”组肿瘤组织相关巨噬细胞microRNA的表达谱的异常变化,使其恢复正常。结论〓加味黄芪建中汤可使“脾气虚”组microRNA的表达谱接近恢复正常。

    Abstract:

    Objective To study the expression of tumor associated macrophages microRNA in lung cancer microenvironment spleen deficiency syndrome and the influence of modified HuangqiJianzhong soup. Methods Using bitter and cold, diarrhea, weary, dieting methods of modeling in 50 mices, after the establishment of mouse spleen deficiency syndrome and Lewis lung cancer model, according to the experimental design of the processing of each group of mice, after modeling Huangqi Jianzhong decoction gavage, the mice were killed off the cervical spine, and tumor tissue was collected. Use MicroRNA gene chip for the detection of variations of normal mice macrophages and three groups of tumor tissue macrophage expression of MicroRNA gene. Results MicroRNA microarray detection of tumor associated macrophages of mice found, Spectrum expression of Spleen Qi deficiency group tumor associated macrophage microRNA(compared with the normal group)aberrant, unlike tumor group, has a unique MicroRNA changes, and modified Huangqi Jianzhong decoction can significantly bidirectional regulate “Spleen Qi Deficiency” abnormal changes in expression profiles of tumor-associated macrophages microRNA tissue to make it back to normal. Conclusion The modified Huangqi Jianzhong decoction can make “Spleen Qi Deficiency” microRNA back to normal.
