    摘要:

    目的〓检测灯盏乙素对乳腺癌MDA-MB-231细胞中的环状RNA(Circular RNA,circRNA)的相对表达量的影响,进一步探索灯盏乙素对肿瘤的作用机制。方法〓采用实时荧光定量PCR的方法,检测对照组与不同剂量组的灯盏乙素处理后的乳腺癌细胞中circRNA OXNAD1与SMARCA5的相对表达量。结果〓灯盏乙素在低剂量(1、10、25μm)的情况下,使得乳腺癌MDA-MB-231细胞中circRNA OXNAD1的表达量显著降低(P<0.05),在高剂量(100μm)时显著升高(P<0.05)。灯盏乙素在低剂量(1、10、25、50μm)的情况下,使得乳腺癌MDA-MB-231细胞中环状RNA SMARCA5的表达量显著降低(P<0.05),在高剂量(100μm)时显著升高(P<0.05)。结论〓灯盏乙素影响circRNA OXNAD1与SMARCA5的表达,并且存在剂量反应关系,低剂量与高剂量呈现出相反的表达量变化。

    Abstract:

    Objective To investigate the effect of scutellarin on the relative expression of circRNA in breast cancer MDA-MB-231 cells, and to further explore the mechanism of scutellarin on tumor. Methods The relative expression of circRNAs OXNAD1 and SMARCA5 in breast cancer cells treated with scutellarin was detected by real-time quantitative PCR in the control group and the different dose groups. Results The relative expression of circRNA OXNAD1 in breast cancer MDA-MB-231 cells was significantly decreased(P<0.05) at low dose(1, 10 and 25μm), and significantly increased at high dose(100μm)(P<0.05). The expression of circRNA SMARCA5 in breast cancer MDA-MB-231 cells was significantly decreased(P<0.05) at the low dose(1, 10, 25, 50μm), and significant at high dose(100μm)(P<0.05). Conclusion Scutellarin has an effect on the expression of circRNA OXNAD 1 and SMARCA5 with a dose-response relationship, the low doses and high dose showed opposite expression changes.
