针刺肾俞、足三里抑制大鼠绝经后骨质疏松症的机制研究*

    摘要:

    目的探讨针刺肾俞足三里调节wnt/β-catenin通路抑制大鼠绝经后骨质疏松症(PMOP)的作用机制。方法将50只雌性SD大鼠随机分为空白组、假模型组、模型组、穴位组和非经非穴组。治疗组、非经非穴组每天针刺1次,留针30 min,10次为1个疗程,疗程间休息5 d,共针刺6个疗程后,X线检测股骨颈、股骨骨密度,比色法检测大鼠血清碱性磷酸酶(ALP)活性及RT-PCR法检测腰椎Wnt1,β-catenin,GSK-3β,Dsh,mRNA表达。结果与模型组、非经非穴组比较,实验组大鼠股骨颈骨密度明显升高,血清ALP活性明显降低,Wnt1、GSK-3β、Dsh的mRNA表达差异均有统计学意义(P<0.05)。结论针刺肾俞、足三里穴对绝经后骨质疏松症有明显抑制作用,其作用机制可能与调节Wnt/β-Catenin通路中信号分子表达有关。

    Abstract:

    Objective To explore the mechanisms of acupuncture at “Shenshu”(BL 23) and “Zusanli”(ST 36) on the wnt/β-catenin pathway in the treatment of postmenopausal osteoporosis(PMOP) in rats. Methods Fifty female SD rats were randomly divided into a blank group, a sham-model group, a model group, a acupoint group and a non-acupoint group. The treatment group and non-acupoint group were given acupuncture once per day, for 30miniter, 10 treatments made one session. There were 5 days at the interval between two sessions. Six sessions of treatment were required. Bone mineral density were measured using a Dual Energy X-Ray Bone Denstormeter. Activity of alkaline phosphatase(ALP) in serum was measured by chromatometry. The expression of Wnt1, glycogen synthase kinase-3β(GSK-3β), β-catenin, Dsh and mRNA in lumbar vertebra were detected by RT-PCR. Results Bone mineral density in the experimental group were higher than those in the model group and non-acupoint group. And serum ALP were lower than those in the model group. The expression of Wnt1, GSK-3β, β-catenin, Dsh and mRNA were significantly difference(P<0.05). Conclusion Acupuncture at “Shenshu”(BL 23) and “Zusanli”(ST 36) possesses marked preventive and therapeutic effect for PMOP. This action is probably associated with the regulation of the key molecules of Wnt/β-catenin signaling pathway.
