Effects of Capparis Spinosa L Combined with Moxibustion on Treg and Thl7-related Cytokines in Rats with Adjuvant Arthritis

    摘要:

    目的观察刺山柑、艾灸和二者联合治疗对佐剂型关节炎大鼠血清IL-17、IL-6,TGF-β、IL-10、和IL-23含量的影响,探讨其调节Th17和Treg相关细胞因子的作用机制。方法60只大鼠随机分为正常组、模型组、阳性药组、刺山柑组、艾灸组和综合组,于大鼠足跖皮下注射0.1 mL弗氏完全佐剂建立AA模型。造模成功后,阳性药组以甲氨蝶呤灌胃,刺山柑组以刺山柑外敷,艾灸组进行艾条温和灸肾俞、足三里,综合组进行联合治疗,各组均治疗3疗程(21 d)。分别在每1个疗程结束后观察大鼠AI指数评分和足爪容积,并用ELISA检测大鼠血清IL-17、IL-6,TGF-β、IL-10、和IL-23含量。结果造模后,大鼠关节肿胀明显,Th17/Treg失衡。经治疗后,各组大鼠足跖炎症情况均有明显改善,尤其是综合组7 d显著疗效。各组干预治疗均能逆转IL-17、IL-6和IL-10水平(P<0.01或P<0.05),仅有刺山柑组和综合组能显著降低IL-23水平(P<0.01或P<0.05),各组TGF-β水平无显著改变。结论刺山柑组和综合组消炎作用迅速,刺山柑、艾灸和综合治疗的方法均能使免疫细胞Th17/Treg达到动态平衡,其中刺山柑和综合治疗更具优势。

    Abstract:

    Objective To observe the effects of Capparis Spinosa L, moxibustion and their combination on serum levels of IL-17、 IL-6、 TGF-β、 IL-10 and IL-23 in AA rats, and to explore the mechanism of their regulation of Th17 and Treg-related cytokines. Methods Sixty rats were randomly divided into normal group, model group, positive drug group, Capparis Spinosa L group, moxibustion group and comprehensive group. AA model was established by subcutaneous injection of 0.1 mL Freund’s complete adjuvant into rat plantar. After the successful establishment of the model, the positive drug group was given methotrexate by gastric lavage, the prickly citrus group was given prickly citrus external application, the moxibustion group was given moxibustion with moxibustion sticks for mild moxibustion of Shenshu and Zusanli, and the comprehensive group was given combined treatment. Each group was treated for 3 courses(21 days). AI index score and foot volume were observed at the end of each course. Serum levels of IL-17、 IL-6、 TGF-β、 IL-10 and IL-23 were measured by ELISA. Results After the establishment of the model, the joint swelling was obvious and Th17/Treg was imbalance. After treatment, the inflammation of rat’s plantar paw in each group was significantly improved, especially in comprehensive group on 7 days. Intervention treatment could reverse the levels of IL-17、 IL-6 and IL-10 in all groups(P<0.01 or P<0.05). Only Capparis Spinosa L group and comprehensive group could significantly reduce the levels of IL-23(P<0.01 or P<0.05). There was no significant change in the levels of TGF-βin all groups. Conclusion The anti-inflammatory effect of Capparis Spinosa L group and comprehensive group is rapid. Th17/Treg of immune cells can be dynamically balanced by the methods of Capparis Spinosa L, moxibustion and comprehensive treatment, among which Capparis Spinosa L and comprehensive treatment are more advantageous.
