Effects of Ningshen Qushi Huayu Decoction on AT1R, HIF-1α and HO-1 mRNA Expression in Lupus Nephritis Mice

    摘要:

    目的观察宁肾祛湿化瘀汤对狼疮肾炎小鼠血管紧张素Ⅱ 1型受体(AT1R)、缺氧诱导因子(HIF-1α)和血红素氧合酶-1(HO-1) mRNA表达的影响。方法 采用慢性移植物抗宿主病(cGVHD)的方法复制狼疮肾炎(LN)小鼠模型,分为空白组、模型组、宁肾祛湿化瘀汤低、中、高剂量组。空白组为无造模及生理盐水干预,模型组生理盐水干预,宁肾祛湿化瘀汤低中高剂量组分别以19.5、39、78 g/kg浓度灌胃干预。干预8周后,处死小鼠,无菌条件下取肾,采用qPCR检测肾组织中AT1R、HIF-1α、HO-1 mRNA表达。结果 造模后第8周,空白组蛋白尿发生率0%,模型组小鼠蛋白尿发生率100%;宁肾祛湿化瘀汤低、中、高剂量组蛋白尿明显低于模型组且存在剂量趋势;肝、肾、肠HE染色结果显示模型组均有明显炎性细胞浸润,ELISA显示血清中IgG、IgG1、IgG2含量明显增加(P<0.05),提示狼疮肾炎小鼠造模成功。与空白组相比,模型组肾组织中AT1R、HIF-1α、HO-1 mRNA表达升高(P<0.05);与模型组比较,宁肾祛湿化瘀汤低、中、高剂量组AT1R、HIF-1α、HO-1 mRNA表达降低(P<0.05),且存在剂量趋势。结论 宁肾祛湿化瘀汤可改善狼疮肾炎小鼠模型症状,其作用机制可能与降低肾脏中AT1R、HIF-1α、HO-1 mRNA表达有关。

    Abstract:

    Objective To observe the effect of Ningshen Qushi Huayu decoction on the expressions of angiotensin II type 1 receptor(AT1R), hypoxia inducible factor 1α(HIF-1α) and heme oxygenase-1(HO-1) in lupus nephritis mice. Methods Lupus nephritis mice model were induced by chronic graft-versus-host disease(cGVHD). The mice were divided into blank group, model group and low, medium and high dose groups of Ningshen Qushi Huayu decoction. In the blank group, there was no model making and normal saline intervention. In the model group, normal saline intervention. In the low, middle and high dose groups, Ningshen Qushi Huayu decoction were administered by gavage at the concentrations of 19.5, 39 and 78 g/kg, respectively. After 8 weeks of intervention, mice were executed and their kidneys were taken under aseptic conditions. The gene expressions of AT1R, HIF-1α and HO-1 in kidney tissues were detected by qPCR. Results The incidence of proteinuria was 0% in the blank group and 50% in the saline group (model group in seven weeks. HE staining of liver, kidney and intestine showed that inflammatory cells infiltrated significantly in the model group. ELISA showed that the contents of IgG, IgG1 and IgG2 in serum increased significantly(P<0.05), suggesting that the model of lupus nephritis was successful in mice. Compared with the blank group, the expressions of AT1R, HIF-1a and HO-1 in saline group increased(P<0.05). Compared with model group, the expressions of AT1R, HIF-1a and HO-1 in low, middle and high dose groups of Ningshen Qushi Huayu decoction were decreased(P<0.05), and there was a dose depend trend. Conclusion Ningshen Qushi Huayu decoction can improve the symptoms of lupus nephritis mouse model, and its mechanism may be related to the decreased expressions of AT1R, HIF-1α, and HO-1 genes in kidney.
