    目的探讨调督针刺对阿尔茨海默病(Alzheimer’s disease,AD)大鼠学习记忆能力的改善作用。方法 将雄性SD大鼠48只,按随机数字表法随机分为4组,即:空白组、假手术组、模型组、针刺组,每组12只。采用双侧海马注射Aβ1-42 5 μL制备AD大鼠模型。造模成功后第8天开始,针刺组取“百会”“风府”针刺,并接通脉冲针灸治疗仪[疏波(频率为2 Hz)]留针30 min,每日治疗1次,连续治疗28 d。采用Morris水迷宫试验检测大鼠学习记忆能力,监测各组大鼠体质量情况,光镜下观察海马病理组织形态学,采用免疫组织化学法检测海马组织白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、胶质纤维酸性蛋白(GFAP)的表达。结果 与模型组比较,针刺组大鼠体质量增长较快(P<0.05);与模型组比较,针刺组大鼠潜伏期明显缩短(P<0.05),游泳距离明显缩短(P<0.05),神经细胞排列较为规则、神经元萎缩较少,IL-6、TNF-α、GFAP表达明显减少(P<0.05)。结论 调督针刺能够改善AD模型大鼠学习、记忆能力,减轻炎症反应,但相关的分子调控机制仍需要进一步研究。

    Abstract:

    Objective To observe the effect of Tiaodu acupuncture on improving the learning and memory ability of Alzheimer’s disease(AD) rats. Methods 48 male SD rats were selected and divided into 4 groups by random number table, including blank group, sham operation group, model group and acupuncture group, each group with 12 rats. AD models were prepared by injecting Aβ1-42 5 μL in both hippocampus. Starting from the 8 d after successfully modeling, the acupuncture group was given acupuncture therapy on the Baihui and Fengfu, and connected the pulse acupuncture treatment instrument [Shubo(frequency: 2 Hz)] for 30 min, once a day, treated for 28 d. Morris water maze was used to test the learning and memory ability of rats and the weight were monitored. The pathomorphology of hippocampus were observed under light microscope and the expressions of IL-6, TNF-α and GFAP were detected by immunohistochemistry. Results Compared with the model group, the weight of rats in acupuncture group increased rapidly(P<0.05), the latency and swimming distance of the acupuncture group were significantly shortened(P<0.05), the arrangement of nerve cells were regularly, and the number of neurons atrophy were less, also the expressions of IL-6, TNF-α, GFAP were significantly reduced(P<0.05). Conclusion Tiaodu acupuncture could improve the learning and memory ability of AD model rats, and reduce the inflammatory response, however the related molecular mechanism need further study.
