    摘要:

    目的研究比较芍药根的栓皮部、韧皮部和木质部3个不同部位的特征图谱差异。方法 通过高效液相色谱法建立特征图谱,采用ODS柱,柱温 30℃,乙腈-0.1%磷酸水溶液梯度洗脱,流速为1.0 mL·min-1,检测波长为230 nm。结果 特征图谱显示:栓皮部分离出15个共有峰,韧皮部分离出13个共有峰,木质部分离出7个共有峰,各部分特征峰易于识别、分离度较好。主成分分析得出3个部分共有的主要成分:芍药苷、芍药内酯苷和苯甲酸。芍药苷在3个部位中的含量无显著差异,芍药内酯苷在栓皮部和韧皮部中含量无明显差异均高于木质部中的含量,苯甲酸在栓皮部的含量高于韧皮部。结论 栓皮部提取液能分离出更多的成分且浓度较高,木质部提取液分离出的成分少且浓度低,韧皮部介于两者之间。

    Abstract:

    Objective To study and compare the differences of characteristic chromatograms regarding 3 parts from root of Paeonia lactiflora Pall.. Methods Establishment of characteristic chromatogram by high performance liquid chromatography (HPLC). ODS column was used in this study, the column temperature was 30 ℃. The gradient elution of acetonitrile -0.1 % phosphoric acid solution was conducted with the flow rate of 1.0 mL·min-1, and the detection wavelength was 230 nm. Results The characteristic chromatogram showed that 15 common peaks were isolated from cork, 13 common peaks isolated from phloem, 7 common peaks isolated from xylem by analyzing within 3 groups. The characteristic peaks of each part are easy to identify and have good resolution. Principal component analysis obtained three main components in common: paeoniflorin, albiflorin and benzoic acid. There was no significant difference in the content of paeoniflorin in the three parts, and the content of albiflorin in the cork and phloem was not significantly different than that in the xylem, the content of benzoic acid in the cork is higher than that in the phloem. Conclusion Cork contains more components with higher content, xylem has the least components and lower content, phloem is somewhere in between.
