Abstract:Objective To observe the effect of Yishen Huazhuo Decoction on the learning and memory ability and psychiatric behavior of senescence-accelerated-prone mouse-8 (SAMP8). Methods Twenty 3-month-old male SAMP8 mice were randomly divided into model group (SAMP8) and Yishen Huazhuo Decoction (YSHZ) group, with 10 mice in each group. Ten 3-month-old male senescence-accelerated mouse resistant 1(SAMR1) mice were used as the control group (SAMR1). YSHZ group was given Yishen Huazhuo Decoction by 6.24 g/(kg·d) of crude drug, while SAMP8 and SAMR1 groups were given the same amount of distilled water once a day for 12 weeks. Morris water maze (MWM) was applied to evaluate the learning and memory ability of the mice. Open field test (OFT) and elevated plus maze (EPM) were employed to measure their psychiatric behavior. Results The results of MWM showed that there was no significant difference in the average swimming speed among the groups (P>0.05). Compared with SAMR1 group, the escape latency of SAMP8 group was significantly prolonged and the number of crossing platform was significantly reduced (P<0.05). Compared with SAMP8 group, the escape latency of YSHZ group was significantly shortened and the number of crossing platform was significantly increased (P<0.05). The results of OFT and EPM revealed that compared with the SAMR1 group, the total movement distance, the number of times to enter the central zone and the proportion of time to stay in the open arms of SAMP8 group were significantly reduced (P<0.05), while compared with SAMP8 group, the first two indicators of YSHZ group were significantly increased (P<0.05), and the proportion was increased (P>0.05). Conclusion Yishen Huazhuo Decoction can effectively improve the early learning and memory impairment and psychiatric disorders of SAMP8 mice.
